The aim of our study was to identify free light chains of immunoglobulin's in potential donors of the bone marrow among of the kirghiz nationality.
MATERIALS AND METHODS:
A total study group comprised 86 voluntary donors from of the National Register of Hematopoietic Stem Cells Kirghizia. Of them from 2004 to 2006 years the explore in the serum blood on content of free light chains of immunoglobulin's at 78 donors of kirghiz nationality in the laboratory of hybridoms technology CNIRRI FASR, St. Petersburg and 8 donors were explore from 2007 to June 2017 years in the laboratory "InterMed", total of women 27 and men 59, in aged 18 to 50 years. The donors lived temporarily in St. Petersburg, Russia.
DEFINITION IMMUNOGLOBULINS TYPES OF HEAVY AND FREE EASY CHAINS IN THE SERUM BLOOD:
The immunoglobulins were typed methods in a direct solid-phase immune -enzyme analysis. Paraproteins were immobilized on a solid phase sera diluted 1:105 and 1: 106 on 0.1 m carbonate-bicarbonate buffers pH 9.5. Heavy chain isotypes and light chain types of myeloma immunoglobulins were determined using conjugant's of monoclonal antibodies (MCAT) with horseradish peroxides. All MCAT were obtained and described in the laboratory of the hybrid technology CNIRRI P FASR, St. Petersburg, the Ministry of Health of Russia.
RESULTS AND DISCUSSION:
The results of our studies among potential voluntary donors who considered themselves to be practically healthy people on content of free easy chains of immunoglobulin's in the serum blood, were detection at the two men had elevated titers IgAk and λ chains and in two men with IgGk and λ chains. At detailed conversation, in the anamnesis at 2 donors the chronic enterocolitis which has transferred a virus hepatitis, all 4 suffer a chronic pyelonephritis, and in the general analysis of urine traces of protein. In detecting such similar diseases, donors need to be excluded from donors bone marrow and included in the data base of the National Registry of Hematopoietic Stem Cells, as excluded donor from donation bone marrow.
Thus, that at donors bone marrow among kirghiz nationalities, high titers IgA, IgG with elevated k and λ-chains were detected in prophylactic studies.
CONCLUSIONS:
1. At inclusion the donor in the National Register of Hematopoietic Stem Cells, a thorough and qualitative selection of the donor is necessary.
2. Explore voluntary donors on indications before inclusion in the National Register of hematopoietic stem cells on the content of free light chains of immunoglobulin's with a prophylactic purpose.
3. It is necessary to investigate donors on the immunoglobulin of heavy and free lungs (k, λ) chains in blood and urine, when planning closely related, unrelated bone marrow transplantation.
4. When detecting high titers at the potential donors of the free light chain, the immunoglobulin's with elevated k, λ chains, should be excluded removed from bone marrow donation and further examination, and observeds in the therapist, nephrologist, hematologist.
